懸浮型概念：培養(yǎng)時(shí)不貼附于底物而呈懸浮狀態(tài)生長來源：自血，脾或骨髓，尤以血中白細(xì)胞癌腫細(xì)胞特點(diǎn)：在懸浮中生長良好細(xì)胞圓形，單個(gè)或小細(xì)胞團(tuán)優(yōu)點(diǎn)：生存空間大，提供數(shù)量大，傳代方便（不需消化），易于收獲，可獲得穩(wěn)定狀態(tài)缺點(diǎn)：觀察不方便，很多細(xì)胞不能懸浮生長（尤以正常細(xì)胞）

懸浮生長細(xì)胞的傳代：直接離心后吸去培養(yǎng)上清液，用新鮮培養(yǎng)液稀釋懸浮細(xì)胞，一般按 1：2-1：5 稀釋細(xì)胞 后，分瓶繼續(xù)培養(yǎng)。

貼壁生長細(xì)胞的傳代：吸去培養(yǎng)液，用 PBS 洗滌細(xì)胞一次，加入消化液，在 37℃中消化約 3～10 分鐘，待細(xì) 胞開始變圓脫落時(shí)，加入新鮮培養(yǎng)液，終止消化液，懸浮和吹打分散細(xì)胞。1000rpm 離心 5 分鐘，去除上清培養(yǎng)液，用新鮮培養(yǎng)液懸浮細(xì)胞。1：2-1：4 稀釋細(xì)胞后，分瓶繼續(xù)培養(yǎng)。MuM－2C細(xì)胞，人眼脈絡(luò)黑色素瘤細(xì)胞EA.hy926細(xì)胞，HUVEC和A549的融合株

MoBiTec provides a wide range of useful vectors for the B. megaterium system that are adaptable to most applications ａnd protein purification methods. These include vectors carrying a secretion signal peptide sequence ａnd a 6xHis-tag.

 

With our new hp-vectors yields up to 10-fold higher compared to the basic plasmids are possible. All plasmids have established multiple cloning sites （MCS） for versatile cloning. Furthermore we offer vectors encoding C- or N-terminal His-tags for easy purification. The protein secretion with LipA or YocH signal peptides is up to nine-fold increased. Induction of protein expression of all vectors is achieved by the tightly regulated ａnd efficiently inducible xylose operon.

Versatile system with a wide range of vectors

The B. megaterium expression system provides a versatile ａnd easy-to-handle tool for stable ａnd high-yield protein production, both small- ａnd large-scale.

 

B. megaterium has proven to be an excellent alternative host to E. coli for heterologous gene expression. Unlike other bacilli strains, proteolytic degradation by alkaline proteases is avoided. In addition, there are no endotoxins found in the cell wall.

B. megaterium protoplasts specifically optimized for transformation

The protoplasts supplied by MoBiTec are from B. megaterium strain WH320, developed by Prof. Dr. W. Hillen at the Institute of Microbiology in Erlangen, Germany. These protoplasts are prepared according to an optimized protocol resulting in the highest transformation efficiencies.

 

For secretion vectors we offer protoplasts of strain MS941. Both strains are mutants of DSM319, where WH320 is a chemically mutant, while MS941 has a defined deletion in the gene of the major extracellular protease NprM.

Cell Specification
The vascular endothelial cells contribute to the maintenance of vascular homeostasis. They synthesize ａnd secrete activators as well as inhibitors of both the coagulation system ａnd the fibrinolysis system, ａnd mediators that influence the adhesion ａnd aggregation of blood platelets. Endothelial cells also release molecules that control cell proliferation ａnd modulate vessel wall tone. Many of these, ａnd other, processes can be studied in vitro using cultured cells, ａnd human umbilical vein ａnd artery derived endothelial cells are the most commonly used cell types for such studies. Except for the common endothelial cell features such as “cobblestone” morphology, positive staining for Factors VIII ａnd the ability to take up acetylated low-density lipoprotein [1], human umbilical endothelial cells also stein positively with CD-31 [2]. Cells pretreated with IL-1 or TNF-α also selectively express E-selectin [3] ａnd VCAM [4].
HUAEC from ScienCell Research Laboratories are isolated from human umbilical artery. HUVEC are cryopreserved at the end of secondary culture ａnd delivered frozen. Each vial contains >5 x 105 cells in 1 ml volume. HUAEC are characterized by immunofluorescent method with antibodies to vWF/Factor VIII ａnd CD31 (P-CAM) ａnd by uptake of DiI-Ac-LDL. HUAEC are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast ａnd fungi. HUAEC are guaranteed to further expand for 15 population doublings at the conditions provided by ScienCell Research Laboratories.

HUAEC are for research use only. It is not approved for human or animal use, or for application in in vitro diagnostic procedures.

Directly ａnd immediately transfer cells from dry ice to liquid nitrogen upon receiving ａnd keep the cells in liquid nitrogen until cell culture needed for experiments.

Instruction for culturing cells
Caution: Cryopreserved cells are very delicate. Thaw the vial in a 37oC waterbath
and return them to culture as quickly as possible with minimal handling!
Set up culture after receiving the order:
1. Prepare a poly-L-lysine coated flask (2 μg/cm2, T-75 flask is recommended) ａnd leave the flask in incubator overnight (minimum one hour at 37oC incubator).
2. Prepare complete medium: decontaminate the external surfaces of medium ａnd medium supplements with 70% ethanol ａnd transfer them to sterile field. Aseptically open each supplement tube ａnd add them to the basal medium with a pipette. Rinse each tube with medium to recover the entire volume.
3. Rinse the poly-L-lysine coated flask with sterile water twice ａnd add 20 ml of complete medium to the flask. Leave the flask in the hood ａnd go to thaw the cells.
4. Place the vial in a 37oC waterbath, hold ａnd rotate the vial gently until the contents are completely thawed. Remove the vial from the waterbath immediately, wipe it dry, rinse the vial with 70% ethanol ａnd transfer it to a sterile field. Remove the cap, being careful not to touch the interior threads with fingers. Using a 1 ml eppendorf pipette gently resuspend the contents of the vial.
5. Dispense the contents of the vial into the equilibrated, poly-L-lysine coated flask. A seeding density of 5,000 cells/cm2 is recommended.
Note: Dilution ａnd centrifugation of cells after thawing are not recommended since these actions are more harmful to the cells than the effect of DMSO residue in the culture. It is also important that endothelial cells are plated in poly-L-lysine coated culture vessels that promote cell attachment.
6. Replace the cap or cover of flask, ａnd gently rock the vessel to distribute the cells evenly. Loosen cap if necessary to permit gas exchange.
7. Return the culture vessels to the incubator.
8. For best result, do not disturb the culture for at least 16 hours after the culture has been initiated. Change the growth medium the next day to remove the residual DMSO ａnd unattached cells, then every other day thereafter. A healthy culture will display cobblestone or spindle shaped morphology, nongranular cytoplasm ａnd the cell number will be double after two to three days in culture.
Maintenance of Culture:
1. Change the medium to fresh supplemented medium the next morning after establishing a culture from cryopreserved cells. For subsequent subcultures, change medium 48 hours after establishing the subculture.
2. Change the medium every other day thereafter, until the culture is approximately 50% confluent.
3. Once the culture reaches 50% confluence, change medium every day until the culture is approximately 80% confluent.
Subculture:
1. Subculture the cells when they are over 90% confluent.
2. Prepare poly-L-lysine coated cell culture flasks (2 μg/cm2).
3. Warm medium, trypsin/EDTA solution, trypsin neutralization solution, ａnd DPBS to room temperature. We do not recommend warming the reagents ａnd medium at 37oC waterbath prior to use.
4. Rinse the cells with DPBS.
5. Incubate cells with 10 ml of trypsin/EDTA solution (in the case of T-75 flask) until 80% of cells are rounded up (monitored with microscope). Add 10 ml of trypsin neutralization solution to the digestion immediately ａnd gently rock the culture vessel.
Note: Use ScienCell Research Laboratories’ trypsin/EDTA solution that is optimized to minimize the killing of the cells by over trypsinization.
6. Harvest ａnd transfer released cells into a 50 ml centrifuge tube. Rinse the flask with another 10 ml of growth medium to collect the residue cells. Examine the flask under microscope to make sure the harvesting is successful by looking at the number of cells left behind. There should be less than 5%.
7. Centrifuge the harvested cell suspension at 1000 rpm for 5 min ａnd resuspend cells in growth medium.
8. Count cells ａnd plate them in a new, poly-L-lysine coated flask with cell density as recommended.
Caution: Handling human derived products is potentially bioharzadous. Although each cell strain testes negative for HIV, HBV ａnd HCV DNA, diagnostic tests are not necessarily 100% accurate, therefore, proper precautions mush be taken to avoid inadvertent exposure. Always wear gloves ａnd safety glasses when working these materials. Never mouth pipette. We recommend following the universal procedures for handling products of human origin as the minimum precaution against contamination [1].
